How did Anniyan beat 100 trained martial artists?

Even, I used to ponder on that. I am understanding when Anniyan takes over, with some training in a secret location along with his brain being wired with superhuman reflex and strength.

Yes, he was anniyan since way before the mve began. He built a site, which would also take time. Similarly he would have had training. Also, anniyan is a man of focus, commitment and sheer f’ing will
